TheFluoroSpotassayisaninventivedevelopmentoftheELISpotassayandusesfluorescentdetectioninsteadofcolorimetric.Thisallowsforsimultaneousdetectionofseveralanalytes,whichisamajoradvantage.ThehumanIL-1β/IL-6/TNF-αFluoroSpotkitisperfectforuserswhowishtohaveaconvenientassaywithminimizedassayvariABIlity.Itenablesenumerationofcellssecretingone,twoorthreeanalytes.Thekitincludeslowfluorescentplatespre-coatedwithmonoclonalcaptureantibodies,monoclonaldetectionantibodies,secondaryreagentsconjugatedtofluorophoresandafluorescenceenhancer.LPSisincludedaspositivecontrolstimulus.

Theassayisrecommendedfortheanalysisofmacrophages,monocytesanddendriticcells.ThesekitshavenotbeenevaluatedforT-cells.

Foranalysis,werecommendtheuseofanautomatedFluoroSpotreaderequippedwithfiltersforexcitation490nm/emission510nm(LED490,FITC)for490-reagentdetection,excitation550nm/emission570nm(LED550,Cy3)for550-reagentdetectionandexcitation640nm/emission660nm(LED640,Cy5)for640-reagentdetection.Filtersshouldhavehighspecificitytoavoidbleed-throughartifacts.

IL-1βInterleukin-1ß(IL-1ßorIL-1F2,alsoknownasInterleukin-1beta,IL-1beta,IL1bandInterleukin-1beta)isaproinflammatorycytokineandinducerofacutephaseresponses.IL-1ßisproducedprimarilybymonocytes,macrophages,anddendriticcellsafterinductionbymicrobes.

IL-6Interleukin6(IL-6,IL6)isapleiotropiccytokinethatisproducedbymanydifferentcelltypesandplaysaroleinawiderangeoffunctions,suchasimmuneresponses,acute-phasereactions,andhematopoiesis.AmongotherthingsitaugmentsantibodyproductionfromactivatedBcellsinvitro.

TNF-αTumornecrosisfactor-α(TNF-α,alsoknownasTNF-alpha,TNF-a,TNFa,andTumornecrosisfactor-alpha)isproducedbymanydifferentcelltypese.g.monocytes,macrophages,TcellsandBcells.AmongthemanyeffectsofTNF-α,areprotectionagainstbacterialinfection,cellgrowthmodulation,immunesystemregulationandinvolvementinsepticshock.
